【择要】Relying on simultaneous absorption of two or more near-infrared photons by a fluorophore, multiphoton microscopy can achieve deep tissue penetration and efficient light detection noninvasively, which makes it very suitable for thick-tissue and in vivo bioimaging. However, most conventional organic fluorophores suffer from severe aggregation-caused quench (ACQ) effect, which means their fluorescence is easily quenched at aggregate or solid state. In 2001, Tang and coworkers observed a diametrically opposite phenomenon called “aggregation-induced emission” (AIE), which provides an ideal solution to the notorious ACQ problem. Based on this mechanism, hydrophobic AIE molecules can be encapsulated with various types of nanoparticles, or form nanoaggregates in an aqueous solution spontaneously. AIE nanoparticles have bright fluorescence, large multiphoton absorption cross-section, and are very resistant to photobleaching even under high-power laser irradiation, which facilitates their applications in long-term multiphoton microscopy as nanoprobes. In this talk, I will introduce some exemplary applications of AIE nanoparticles in two- and three-photon in vivo functional brain imaging of mice, such as brain-microglia imaging, 3D architecture reconstruction of brain blood vessel, and blood velocity measurement. We also applied AIE nanoparticles in three-photon imaging of zebrafish at different growth stages.

 

【陈诉人简介】钱骏,博士,男,浙江大学光电迷信与工程学院传授,博士生导师。20046月在浙江大学光电系获学士学位,20096月在浙江大学光电系获博士学位,20097月至20116月在浙江大学光及电磁波研讨中央从事博士后研讨,20116月至201112月在浙江大学光电系担当讲师,20121月至201512月在光电系担当副传授,20121月成为浙江大学求是青年学者、担当博士生导师,20161月至今在光电学院担当传授。2006年至2007年曾在美国纽约州立大学布法罗分校“激光、光子学、生物光子学研讨中央”停止学术拜访,师从纳米生物光子学的国际领武士物Paras N. Prasad传授。次要从事纳米生物光子学的研讨。已发布SCI 论文50多篇,此中,以第一/通讯著作人身份在Adv. Mater.Angew Chem Int EditACS Nano, Biomaterials, Nano Research, Theranostics, Optics. Lett.,Biomed. Opt. Express., J. Biomed. Opt.等高程度期刊上发布论文30多篇。著有英文学术专著一章节。承当了国度天然迷信基金(面上和青年),973863子课题,浙江省出色青年迷信基金等项目。
